FBLD Workflow ______________________________________
At NMX, our workflow and methodologies have been optimized to overcome common pitfalls within the FBLD process, which often results in costly and time-consuming limitations. Between the stages of “initial screening” and “confirmed weak binders”, common problems arise which make this process challenging: issues with physico-chemical properties, poor solution behavior resulting in false positives/negatives, high concentration requirements, among others.

NMX’s multifaceted, cyclic approach re-envisions the linear workflow of classic FBLD. We comprehensively study the target and ligand systems early-on to understand the relevant properties and behavior, ultimately avoiding many of the sneaky issues previously mentioned. This allows for quicker progress towards successful compound screening and analoging by NMR, ultimately establishing affinity rankings and structure-activity-relationships, which are crucial pieces of information when moving into bio-assay testing and determining strong binders for leads.
_________ NMR Optimization and Crystallography Services
We understand that many clients struggle with the demands of producing NMR-suitable samples, often having to work with expensive deuterated buffers, being unable to prepare samples in the necessary buffer conditions or unable to remove components critical for sample stability. Recently, NMX has dedicated significant efforts to enable our fragment-based NMR methodologies to work entirely with non-deuterated components, increasing sample viability while improving signal resolution, allowing us to pass the savings directly back to our clients!
Common buffers and components such as HEPES, Tris, PEG, and urea have been successfully optimized for use in their protonated forms with our improved solvent suppression protocols, while more are in-development. NMX can additionally customize protocols for client needs if required buffer conditions fall outside of the currently optimized non-deuterated systems.
Along with continually improving NMR methodologies and offerings for our clients, NMX is excited to offer crystallography services to help elucidate structural and binding site information for client targets. Crystal growth, data analysis, and data refinement can now be completed in-house by our experts, providing greater insights at various points of the FBLD process, particularly in the fragment growth stages within the med chem cycle.
